依托咪酯削弱去甲肾上腺素对自发性高血压大鼠离体胸主动脉收缩作用的实验研究
Study on the Effect Of Etomidate on Attenuating Norepinephrine Induced Contraction of Isolated Thoracic Aorta in Spontaneous Hypertensive Rats

Abstract
OBJECTIVE: To observe the effect of different concentrations of etomidate on contraction of isolated aorta in spontaneous hypertensive rats (SHR) and normotensive rats (NTR). METHODS: Male NTR and SHR, each of 18, were assigned into etomidate low-dose, medium-dose and high-dose intervention groups (5×10-6 mol·L-1, 3×10-5 mol· L-1, 5×10-5 mol· L-1, n=6),normal saline control group (n=6) and solvent control group (20% lipofundin medium-chain triglyceride/long-chain triglyceride (MCT/LCT), n=6). Thoracic aortic rings were isolated and cultured in different solutions, and put into different concentrations of norepinephrine (NE) for contraction. The vascular tension changes (g) of samples were recorded after hanging on the relevant apparatus to observe the range of contraction. RESULTS: Compared with NTR groups, the response of thoracic aorta to NE-induced vascular contraction in SHR groups increased significantly (P<0.05 or P<0.01 ). Compared with control group, medium-dose and high-dose etomidate attenuated vascular contraction in NTR groups (P<0.05 or P<0.01). 3 doses of etomidate attenuated vascular contraction in SHR groups (P<0.05 or P<0.01 ) in dose-dependent manner. Under same concentration of NE, high-dose of etomidate had significantly attenuated vascular contraction in SHR group, compared with NTR group (P<0.05 or P<0.01). CONCLUSION: Compared with NTR, the response of thoracic aorta of SHR to NE increases significantly. Etomidate has significant effect on attenuating the NE-induced contraction of thoracic aorta, especially in SHR.关键词
