Effect of Vascular Endothelial Growth Factor on the Expressions of Ihh and p38 MARK in Osteoblasts

Abstract
Objective To explore the effect of vascular endothelial growth factor (VEGF) on the expressions of Indian hedgehog (Ihh)and p38 mitogen-activated protein kinase (MAPK) in osteoblasts. Methods The osteoblasts from the calvarial of neonatal rats were cultured and treated with different concentrations (0,2,and 20 ng/ml) of VEGF for various durations (0, 12,and 24 hours). The expressions levels of Ihh and its receptor Ptch1 and p38 MAPK in the osteoblasts were determined with real-time polymerase chain reaction. Results Ihh,Ptch1,and p38 MAPK expression were observed in the osteoblasts. With the increase of the concentration of VEGF,the expressions levels of Ihh and p38 MAPK significantly increased. Compared with control group, the expresions of Ihh and p38 MAPK tended to increase with the prolongation of the duration of VEGF treatemnt. Conclusion Ihh and Ptch1 were coexpressed in the osteoblasts. Ihh regulates the osteoblasts in an autocrine manner. VEGF might upregulates the expressions of Ihh by regulating p38 MAPK expression. VEGF-p38 MAPK-Ihh pathway might exist in the osteoblasts.关键词
