固相萃取高效液相色谱法测定万古霉素血药浓度

摘要
Abstract
Objective To establish a HPLC method for determination the concentration of vancomycin in serum. Methods The analysis was conducted with Hypersil C18 column (4. 6 mm × 200. 0 mm, 10 μm).Acetonitrile -potassium dihydrogen phosphate (9:91 ) was served as mobile phase, the flow number was 0. 8 mL · min-1, and the detecting wavelengh was at 236 nm. Serum sample was applied to solid phase extraction(SPE) column, and the eluting agent was 10% acetic acid colonial spirit solution. The eluant was collected and was dried in 65 ℃ water bath with nitrogen gas. Result The linear range of vancomycin in serum was 0. 92 ~ 117. 28 mg · L-1. The recovery rate of low concentration, moderate concentration and high concentration (4. 69,23.45,46. 90 mg · L-1 ) was 100. 92%, 101.48%, 104. 31%, respectively. Both of the intra - day and inter - day RSD were less than 5 %. Conclusion This method is accurate, sensitive and simple. And it is suitable for determination the concentration of vancomycin in serum and the pharmacokinetic study of vancomycin.关键词
