猪瘟病毒GSLZ株的分离与鉴定
Isolation and identification of classical swine fever virus GSLZ strain

Abstract
Samples were collected from pigs with suspected classical swine fever on a pig farm in Lanzhou,Gansu Province,and the E2 gene of classical swine fever virus(CSFV) was amplified by RT-PCR.PK-15 cell lines were inoculated with positive specimens,followed by blind passage culture for 12 generations,the E2 gene was detected from cell cultures of the second generation,typical virions of 25 nm were observed under electron microscopy,and named Gansu Lanzhou(GSLZ)strain. Sequence analysis showed that the nucleotide sequences of E2 gene had 83.1 % to 99.8% homology between the reference strains and isolated strains. However,the predicted antigenic epitope was drifted compared with the Chinese C strain.Rabbits infected with PK-15 cells cultures of the 8th generation did not show any body temperature change,while piglets showed typical clinical symptoms and pathological changes. These results indicated that the CSFV GSLZ strain was a virulent strain which had strong virulence to piglets,whereas it could induce immune responses rather than infection to rabbits.关键词
