Abstract
Objective:To observe the changes of the expressions of MMP-2 and E-cad before and after Forskolin treating the Human Choriocarcinoma BeWo cell line,to investigate the relationship of the fusion status of Choriocarcinoma calls and their invasive and metastatic capabiligy. Methods: BeWo cell lines were cultured and passaged, then were treated by 100mmol/L Forskolin. Syncytin mRNA was detected by RT-PCR, also the changes of the protein, MMP-2 and E-cad were checked by Westem blot. Results:According to the shape and brightness of PCR and Western bands, Syncytin mRNA and E-cad, MMP-2 protein showed time-dependent changes. Along with the reaction of Forskolin, the expression of Syncytin mRNA and E-cad protein increased steadily, while the expression of MMP-2 protein decreased, the differences were significant ( P ＜ 0. 05). Conclusions:The high expression of syncytin might be related with the changes of invasive or metastatic capability post fusion of choriocarcinoma cells. So Syncytin might be a valueble prognostic marker and offer a new treatment ragimen for trophoblastic tumor.关键词
