染料木素对异丙肾上腺素诱导的心肌肥厚大鼠血管活性物质的影响
Effect of genistein on vasoactive substances in myocardial hypertrophy rats induced by isoprenaline

Abstract
OBJECTIVE To explore the protection of genistein (Gen) against rats' myocardial hypertrophy induced by isoprenaline (Iso) and the role of vasoactive substances. METHODS Iso 1 mg· kg-1 was given sc for the settlement of a myocardial hypertrophy model. From the second day on, Gen 0.03, 0.1 and 0.3 μmol·kg -1 was sc given, 30 min after Iso, for 14 d. Twelve hours after the last administration, the indexes of the left ventricle were calculated, the concentration of angiotensin Ⅱ(AngⅡ), endothelin(ET) and atrial natriuretic peptide (ANP)in plasma was investigated with radioimmunity kits. RESULTS Compared with normal control group, indexes of the left ventricle of Iso model group increased from (1.87 ±0.09) to (2.48 ±0. 11) mg·kg-1(P <0.01), AngⅡ content in plasma from (245 ±74) to (354 ±51 ) ng·L-1 (P <0.05), ET from (37 ± 18) to (66 ±20)ng·L-1(P <0.05) , while ANP decreased from (506 ±115) to (265 ± 126) ng·L-1 (P < 0.05 ). Compared with Iso model group, the indexes of the left ventricle in Gen 0.1 and 0.3 μmol·kg-1 groups significantly decreased to 2.28 ±0.10 and (2.25 ±0. 13) mg·kg-1, respectively (P < 0.05 ). Ang Ⅱ content in Gen 0.03, 0.1 and 1 μmol· kg-1 groups statistically decreased to 242 ±77, 198 ± 40 and ( 196 ± 53 ) ng·L-1 with ( P < 0.05 ), respectively. ET content in Gen 0.03 and 0.1 μmol·kg-1 groups significantly decreased to 36 ± 18 and (38 ±9 ) ng·L-1 (P <0.05). ANP content in Gen 0.03, 0. 1 and 0.3 μmol·kg-1 groups significantly increased to 695 ± 177,574 ±219 and (544 ±231) ng·L-1(P<0.05), respectively. CONCLUSION Gen has a protective effect on rats' myocardial hypertrophy induced by Iso, which might be related to the modulation of the balance of vasoactive substances, such as Ang Ⅱ, ANP and ET.关键词
