Abstract
Objective :To study the modulation of L - mevalonate ( L - MVA) on the effects of simvastatin on PTEN expression in cultured rat cardiac myocytes. Methods: Image analysis system and [ 3 H ] - leucine incorporation were used to measure cell surface area and protein synthesis. The mRNA expression of atrial natriuretic peptide (ANP)was determined with RT- PCR. RT- PCR and Western blot were applied to study the mRNA and protein expression of PTEN, respectively. Results: Cell surface area, [3H] -leucine incorporation rate, ANP mRNA levels in simvastatin + L- MVA group were significantly higher than those of simvastatin group (P ＜0.01 ), while the mRNA and protein levels of PTEN in simvastatin + L - MVA group were significantly lower than those of simvastatin group (P ＜0.01 ). Conclusion:L- MVA can decrease the expression level of PTEN, thus reverse the inhibition effects of simvastatin on cardiac myocytes hypertrophy.关键词
