摘要

Abstract

Objective To explore demethylation effect of 5-Aza-2'deoxycytidine (5-Aza-CdR)in gastric cancer cell line AGS and study its clinical applications.Methods Bisulfite sequencing PCR (BSP) and reverse transcriptase PCR ( RT-PCR ) were used to detect CHFR gene promoter's methylation status and its mRNA expression pre and post 5-Aza-CdR treatment in gastric cancer cell line AGS respectively.Results CHFR gene promoter displayed hypermethylated in gastric cancer cell line AGS (methylation rate≥60% ), while it was low after 5-Aza-CdR treatment (methylation rate≤20% );CHFR mRNA expression recovered after 5-Aza-CdR's demethylation.Conclusions CHFR gene's promoter display hypermethylation status in gastric cancer cell line AGS, 5-Aza-CdR can recovery abnormal hypermethylation and induce its mRNA expression significantly.It may be a new idea for the treatment of gastric cancer.
