T-2毒素中毒对低硒大鼠关节软骨细胞凋亡相关基因mRNA表达的影响
Effects of T-2 toxin on the mRNA expression of apoptosis-related gene in articular chondrocytes of selenium-deficiency rats

Abstract
Objective To study the effects of T-2 toxin on the mRNA expression of apoptosis-related gene in articular chondrocytes of selenium-deficiency rats for providing some evidence for understanding the mechanisms leading to chondrocyte apoptosis and the cause of Kashin-Beck disease. Methods Weaning male SD rats were randomly divided into four groups, i.e., normal diet group (Group A), selenium-deficient diet group (Group B),normal diet plus T-2 toxin group (Group C), and selenium-deficient diet plus T-2 toxin group (Group D). And then the rats in Groups A and C were fed with normal diet and those in Groups B and D were fed with seleniumdeficiency diet for 30 d. After that, rats in Groups C and D received an intragastric administration of T-2 toxin (200 ng/(g · d · bw) for 30 d, respectively. The articular cartilage was removed from rats and RNA was extracted from the articular cartilage by Trizol. Real time-PCR was used to determine the mRNA expressions of Bax, Bcl-2 and P53 and caspase-3. Results Compared with that of normal diet rats (73.92 ± 30.01)ng/mL, the blood selenium content was ( 4.16 ± 3.56 ) ng/mL in rats on selenium-deficient diet for 30 d, with a significant difference (P<0.05). Compared to that in normal diet group, the mRNA expressions of P53, caspase-3, Bax and Bcl-2 were up-regulated in selenium-deficient diet group, normal diet plus T-2 toxin group and selenium-deficient diet plus T-2 toxin group. However, the mRNA expression of Bcl-2 was down-regulated, with a significant difference (P<0.05). Compared to that in selenium-deficient diet group, there were up-regulated mRNA expressions of P53,caspase-3 and Bax in selenium-deficient diet plus T-2 toxin group, but down-regulated mRNA expression of Bcl-2;the differences showed significance (P < 0.05). In selenium-deficient diet plus T-2 toxin group, the mRNA expressions of P53, caspase-3 and Bax were higher than those in normal diet plus T-2 toxin group, but the mRNA expression of Bcl-2 was significantly lower than that in normal diet plus T-2 toxin group (P<0.05). Conclusion T-2 toxin (200 ng/(g · bw · d) can cause the mRNA expression change of chondrocytes apoptotic factors in rats on selenium-deficient diet.关键词
