Expression of TFF3 in different positions of the spleen during gastric ulcer in rats

Abstract
Objective To study the expression of trefoil factor 3 (TFF3) in different positions of the spleen during gastric ulcer in rats.Methods Acetic acid-induced ulcer was induced by injecting acetic acid into submucosa of gastric anterior wall.The expression of TFF3 in the spleen of 6 normal rats and 42 rats with gastric ulcer was detected by immunohistochemistry.Results ① In gastric ulcer rats, the spleen was swollen and congested, and the area of white pulp (WP) was enlarged obviously (P<0.05 or P<0.01), which was the largest in the 6th-day group; the marginal zone (MZ) was thickened (P<0.05 or P<0.01), which was the thickest in the 10th-day group, but the area of red pulp (RP) was reduced.② The positive products of TFF3 were brown and yellow granules distributed in the cytoplasm.③ The expression of TFF3 in the spleen was weakly positive in normal group,but was obviously increased in gastric ulcer group (P<0.05 or P<0.01).The TFF3 average optical density (AOD)values in WP were obviously higher in each gastric ulcer group after gastric ulcer than in normal group (P<0.05),which was the highest in the 10th-day group.The TFF3 AOD values in MZ were obviously higher in the 1st-day,2nd-day and 6th-day groups than in normal group (P<0.05 or P<0.01), which was the highest in the 6th-day group.The TFF3 AOD values in RP were obviously higher in the 1st-day, 2nd-day, 6th-day and 14th-day groups than in normal group (P<0.05 or P<0.01), which was the highest in the 6th-day group.④ The expression of TFF3 in MZ of gastric ulcer group had a negative correlation with injury time (P<0.05).Conclusion TFF3 is highly expressed in different regions of the spleen during gastric ulcer in rats.Healing of gastric ulcer may be promoted by enhancing spleen immunoreactivity.关键词
