摘要

Abstract

Objective To investigate the urinary metabonomic variation in diabetic rats in order to explore the application of metabonomics in research of diabetes. Methods Male Sprague-Dawley rats were fed with high-fat and high-sucrose diet for 6 weeks and then injected with vehicle or streptozotocin (STZ) in a dose of 37 mg/kg. Fasting blood glucose (FBG) was monitored weekly after STZ injection. Urines of normal rats and model rats were collected at 1, 2, 4, 6, 8 weeks after STZ injection. Serum insulin (INS), triglyceride (TG), total cholesterol (TC) and free fatty acids (FFA) were measured at 8 weeks after STZ injection and the rats were sacrificed. The metabolic profiles were analyzed using ethyl chloroformate (ECF)derivatization and gas chromatography-mass spectrometry (GC/MS). Results Compared with the control rats, the FBG, TG, FFA were significantly higher while the INS was reduced in the model rats. The metabolism profiles of the model rats were significantly different from that of the control rats, and changed with time. Comparative study was applied and ten biological markers were different between the normal and model rats. Conclusions Significant changes in urine metabonomics occurre in type 2 diabetic rats induced by high-fat and high-sucrose diet and low dose of STZ,and it may reflect to a certain extent the pathological changes of type 2 diabetic rats.
