Abstract

Objective To investigate the effect of curcumin on bronchopulmonary dysplasia induced by hyperoxia in neonatal rats and explore its potential mechanism.Methods Eighty SD newborn rats were randomly divided into 4 groups: room air group, hyperoxia group, hyperoxia + vehicle group, hyperoxia + curcumin group.Pathomorphology of the lungs was observed with hematoxylin-eosine staining.Cell apoptosis in the lung tissue was detected by TUNEL staining.The expression of activated caspase-3 was detected by immunohistochemistry and Western blotting.Results Compared with the normoxic group, rats exposed to hyperoxia showed a histological pattern of alveolar simplification characterized by the presence of larger and fewer distal air spaces.In addition, the radial alveolar counting ( RAC ) was significantly lower and attenuated with curcumin treatment for 14 days.Hyperoxia exposure also resulted in significant cell apoptosis, which was reduced after treatment with curcumin.Both immunohistochemistry and Western blot demonstrated an increased expression of activated caspase-3 in the lung tissue after hyperoxia exposure, which was reduced by curcumin treatment for 4 days.Conclusion Curcumin alleviates bronchopulmonary dysplasia induced by hyperoxia in neonatal rats, probably via antiapoptotic mechanisms.
