黄蜀葵花器官中β-半乳糖苷酶的提取纯化和酶学特性研究
Purification of β-galactosidase from Flowers of Abelmoschus manihot and Its Characteristics

Abstract
In this paper, the orthogonal experiment was used to optimize the condition for the extraction of β -galactosidase from the flowers of Abelmoschus manihot by taking the total activity of the enzyme as index, the saturation of ammonium sulfate in saltingout method was determined based on the enzyme activity and protein content, and the optimum pH -value and temperature as well as the thermal stability for the extracted β - galactosidase were studied by using ONPG as the substrate. The results indicated that the optimum condition of extracting β - galactosidase from the flowers of Abelmoschus manihot was extracting 12 h in 3 - times volume ( V/ W) of phosphate buffer at 4 ℃. The optimum saturation of ammonium sulfate for salting - out was 30% and 70%. The β - galactosidase reached the highest total activity and had better thermal stability at 55 ℃, and its activity decreased rapidly above 70 ℃.关键词
