异补骨脂素对小鼠胚胎前成骨细胞胶原的影响
Effect of Isopsoralen on Osteoporosis Collagen of Mouse Preosteoblasts

Abstract
Objective To evaluate the impact of different concentrations of isopsoralcn on mouse preosteoblasts (MC3T3-E1) proliferation, type Ⅰ collagen and c-Jun subunit of AP-1 transcription factor,and to describe the relation with primary osteoporosis.Methods Cells were divided into normal control group and different concentrations of isopsoralen groups.Cells proliferation were detected by MTT.The content of cells layer collagen were detected by sirius red method.Western-Blot was used to detect the c-Jun subunit of AP-1.RT-PCR was used to detect mRNA expression of type Ⅰ collagen.Results Different concentrations of isopsoralen stimulated groups promoted the proliferation of MC3T3-E1 cells, cells layer collagen content were increased, the expression of c-Jun protein and type Ⅰ collagen mRNA were increased, and all the differences were statistically significant (P＜0.05).Conclusion Isopsoralen may has the mechanism of promoting the expression of nuclear transcription factor C-jun and then stimulating bone collagen synthesis, thus have the function of prevention and treatment of osteoporosis.关键词
