内吗啡肽对小鼠树突状细胞免疫功能的影响
Effect of endomorphins on immunologic functions of mouse bone marrow-derived dendritic cells

Abstract
Aim The aim of the present study was to investigate the effect of endomorphins on the immunologic functions of mouse bone marrow-derived dendritic cells. Methods Bone marrow dendritic cells were extracted from normal, 7 ~ 8-week-old C57BL/6J mice,purified with anti-CD11c ( a specific marker for dendritic cells ) antibody-coated magnetic beads and cultured in the presence of GM-CSF. Then the immature DCs were cultured in the presence of LPS or LPS plus different concentration of endomorphins. DCs were analyzed for expression of the surface markers by flow cytometry. The effect of endomorphins on the DCs chemotaxis was investigated. 3 H-thymidine incorporation was used to detect the proliferation of T cells under the condition of co-culture of T lymphocytes with endomorphin-treated dendritic cells. Results Endomorphins inhibited DCs chemotaxis. Consistently endomorphinstreated dendritic cells inhibited the proliferation of T lymphocytes. All the above-mentioned effects of endomorphins could be reversed by CTOP ( specific mu-opioid receptor antagonist ), suggesting that these effects of endomorphins were mediated by mu-opioid receptors. Conclusion Endomorphins modulate the functional properties of dendritic cells and immune response by acting on mu-opioid receptors.关键词
