摘要

Abstract

Objective To study the liver and kidney toxicities on rats after long-term intravenous injection of Cyclovirobuxine D and PEGylated Cyclovirobuxine D, and to investigate the effect of PEGylation in reducing the toxicity. Methods Fifty Sprague-Dawley rats were randomly divided into 5 groups (10 in each group): the control group, the Cyclovirobuxine D group, the high-, middle- and low-dose groups of PEGylated Cyclovirobuxine D. Body weight, serum biochemical parameters and pathological examination in liver and kidney tissue were examined after intravenous injection of different dosages of Cyclovirobuxine D and PEGylated CyclovirobuxineD for 28 days. Results Blood uric acid level in Cyclovirobuxine D group was much higher than that in control group; there wasn't difference between the PEGylated Cyclovirobuxine D groups and the control group. It suggested that renal function might be affected in Cyclovirobuxine D group. Liver tissue of Cyclovirobuxine D group and PEGylated Cyclovirobuxine D group had pathological change. Liver toxicity of the Cyclovirobuxine D group was higher than that of the PEGylated Cyclovirobuxine D group at the same dose. Conclusion PEGylated Cyclovirobuxine D has lower liver and kidney toxicities than Cyclovirobuxine D.
