摘要

Abstract

Objective To synthesize a new bismuth porphyrin and investigate its interaction with calf thymus DNA ( CT-DNA ). Methods A new bismuth porphyrin Bi [ MPyTMOPP ] NO3 was synthesized and characterized by elemental analysis,1H-NMR, MS spectra (ESI-MS), IR and UV-Vis spectroscopy. The interaction of Bi[ MPyTMOPP] NO3 and CT-DNA was investigated by UV-Vis spectroscopy,fluorescence spectroscopy, circular dichroism (CD) spectroscopy, thermal denaturalization and viscosity experiments.Results The ESI-MS spectra of Bi [ MPyTMOPP ] NO3 had a ionic peak contributed to ( M + H) + at 912.47;In the Sorer band of UV-Vis spectra, a peak with minor hypochromism and bathochromism (reaching 13.7% and 3 nm) was observed,and the intrinsic binding constant was determined to be 8.6 × 103 L/mol. A positively induced CD band appeared in the Soret band of Bi[ MPyTMOPP] NO3 with DNA.The melting temperature of CT-DNA increased by 2.1 ℃ ,and the relative viscosity of CT-DNA didn't vary with the concentration of Bi[ MPyTMOPP] NO3. Conclusion The results suggest that Bi[ MPyTMOPP] NO3 may interact with CT-DNA by outside groove binding.
