Abstract

In order to provide stable and reliable microalgal source for the biofuel production from large scale micmalgae culture , cryopreservation of Botryococcus braunii was investgated and a fast and effective method of cryopreservation was estabjished . Five cryoprotectants (din ethyl sulphoxide , DM SO ; methanol, M eO H ; ethylene glycol, EG ; propylene glycol, PG ; and glycerol, G ly ) at different concentrations were evaluated to determ ine their toxicity and post-thaw viability for B . braunii. The results showed that the optinal post-thaw viability of 98 .9% was achieved at 6% of M eO H . Different cooling procedures were also studied and it indicated that the program med cooling was essenrial for cryopreservation of B . braunii. Culture age of B . braunii affected its post-thaw viability significantly . It was suggested that the microalgae harvested from late exponential phase of cultue was beneficial for cryopreservalion of B . braunii.
