PEG模拟干旱胁迫对石斛DNA表观遗传变化的MSAP分析
MSAP ANALYSIS OF EPIGENETIC CHANGES IN Dendrobium huoshanense UNDER PEG SIMULATED DROUGHT STRESS

Abstract
The methylation-sensitive amplified polymorphism (MSAP) techniques were used to assess genetic and epigenetic variations of Dendrobium huoshanense under drought stress. Results showed that the level of global DNA methylation decreased with increasing of PEG-6000 concentrations. When the tube seedlings were treated with 5% ,10%, 15% of PEG-6000, the levels of DNA methylation dropped to 34.7%, 32.9% and 30.4%, respectively,compared with that for the control group ( 37. 2% ). There was a significantly negative correlation ( r = - 0. 998 )between PEG-6000 and DNA methylation. In contrast with the control, under stress of 5%, 10% and 15% PEG-6000,methylation and demethylation of DNA were 12. 02%, 12. 50%, 15.53% and 8.39%, 8.55% , 8.45%, respectively.The ratios of DNA methylation and demethylation, the polymorphism of DNA methylation increased gradually,accompanied with the enhancement of drought stress.关键词
