摘要
Abstract
Objective Discuss the chemotaxis of vascular endothelial cells incubated by the serum of Patients with severe preeclampsia on the umbilical cord mesenchymal stem cells. Methods Between Oct. 2009 and Nov 2009 at China Medical Uiniversity Affiliated Shengjing Hospital human umbilical vein endothelial cells(HUVEC) were planted in the six-well cluster dishes and incubated with DMEM with different serum( 15％ serum of patients with severe preeclampsia; 15％ serum of healthy pregnant women ;serum-free)for 24 hours. collected the culture solution in a sterile EP, centrifuged the solution for 20 minutes, then added the solution into the lower chamber and the upper chamber was filled with the fifth passage of UC-MSCs ( 1 × 105/mL). Mter 24h of incubation in a 5％ CO2 atmosphere at 37℃,the invasive cells of the lower surface were fixed in 4％ paraformaldehyde solution for 5 min at room temperature. Then the cells were stained with tetrabromofluorescein. The extent of invasion was determined by counting the number of stained cells on the membranes in five randomly selected, non-overlapping fields at 20 magnification under a light microscope. Results The number of stained cells on the membranes of N group:40.87 ± 6.33; the number of stained cells on the membranes of H group :45.47 ± 6.89; the number of stained cells on the membranes of P group :56.07 ± 6.76. P group is the most, The differences were statistically significant(P ＜ 0. 01 ,F = 20.53). Conclusion MSCs has the chemotactic characteristic to vascular endothelial cells incubated by the serum of patients with severe preeclampsia.关键词
