白羽王鸽部分生殖激素表达的发育性变化及其表达的组织差异初探
Studies on Expression Pattern and Regulation Mechanism of Related Genes about Broodiness and Incubation of White King Pigeon (Columba liva )

Abstract
The aim of this study was to test special broodiness and incubation mechanism of pigeon. The experiment was conducted to investigate changes of serum concentrations of PRL and E2 , and the mRNA expression of PRLR and VIP in brain, liver and kidney in male and female pigeon were assayed during the juvenescent phase, broodiness phase and incubation phase by radioimmunoassay (RIA) and real-time PCR. The results showed that, during detection time, the serum concentrations of PRL and E2 in male pigeon were lower than that in female. In adolescence pre-determination, PRL and E2 levels in the male doves did not change significantly. A downward trend of PRL concentration was showed and E2 concentration in female doves significantly increased in later period(124 d). In reproduction cycle, the concentrations change trend of serum PRL were very similar in male and female pigeons. The serum PRL concentrations in broodiness phase were significantly higher than that in adolescence phase and the expression peak of PRLR occurred in incubation phase. With the passage of time, a down-regulation of the serum E2 concentration was showed, and its concentration increased in mid-incubation phase. The two genes were expressed in three organizations (brain, liver, kidney). In brain, the expression level of PRLR in male and female pigeons during the broody period were significantly higher than that during the juvenescent phase. In the first day of incubation, the expression of PRLR was expressed maximum. The expression of VIP in brain during the broodiness period were significantly higher than that during the juvenescent and incubation periods. The results suggest that PRL plays an important role in maintaining broodiness behavior of pigeon.关键词
