Effects of TGF-β on the proliferation of human Th17 cells

Abstract
Objective To investigate the immunoregulatory function of transforming growth factor -β(TGF-β) on human Th17 cells.Methods Human peripheral blood CD+4 T cells purified by anti-CD4 monoclonal antibody conjugated microbeads from healthy donors were randomly divided into observed group and control group, then both groups were stimulated with 5 μg/ml PHA and 5 ng/ml of recombinant human IL-2 in IMDM medium for 4 days at 37 ℃, with 5% CO2, the observed group was given human recombined TGF-β( 100 IU/ml) at the same time.The concentrations of IL-17 protein in the supematants were assayed by enzyme-linked immunosorbentassay (ELISA), and quantity of Th17 cells were detected by flow cytometry.Results The level of IL-17 protein in the supematants of CD4+ T cells in the observed group and control group was (6.11 ±9.20), (15.07 ± 12.70)pg/ml, the observed group was lower (P=0.07); percentage of Th17 cells in the observed group and control group was 2.45% ± 0.62% and 2.81% ± 0.44%, the observed group was significantly lower (P < 0.05).Conclusion TGF-β exerts immunoregulatory function on human Thl7 cells by inhibiting its proliferation in vitro.关键词
