Abstract

Based on the EST sequences from a differentially expressed eDNA library of Sesuvium portulacastrum roots, the full-length eDNA of a late embryogenesis abundant pnotein gene designated as SpLea5 was cloned from S. portulacastrum roots. Sequence analysis showed that the SpLea5 eDNA contained 685 bp with an open reading frame (ORF) of 294 bp and encoded 97 amino acid residues. The deduced amino acid sequence shared the identity of 54%, 52%, 48%, 47%, 47%,46%, 46%, 44% and 41% with that from Tamarix androssowii, Populus suaveolens, Nicotiana tabacum, Citrus sinensis,Citrus unshiu, Jatropha curcas, Gossypium hirsutum, Glycine max and Ricinus communis, respectively. In phylogenetic tree the SpLea5 and other plant-derived Leas were grouped into one large cluster. Semi-quantitative RT-PCR analysis revealed that the SPLea5 was constitutively expressed in all tested organs but at different levels. The SpLea5 was strongly expressed in roots, but weakly in leaves and stems. The expression of SpLea5 was strongly regulated by seawater, NaCI, PEG and ABA, suggesting that the SpLea5 in S.portulacastrum may be involved in responses to salt stress and drought.
