Abstract

The binding reaction of Isoniazid with bovine serum albumin was studied at 292K and 3 11K by fluorescence spectra.It was shown that Isoniazid has a strong ability of quenching the fluorescence of BSA, and the quenching mechanism of Isoniazid to BSA was a dynamic quenching.According to the Forster theory of non-radiation energy transfer, the binding distances (r) was 5.60 nm, the binding constants (Kb) were 5.32l×104 L·mol-1(292K), 3.189×104L·mol-1 (311K),respectively.The thermodynamic parameters ArHm=-20.343 kJ·mol-1,△rSm=20.8 J·mol-1 showed that the interaction of Isoniazid and BSA is mainly driven by electrostatic force.Synchronous spectrum was used to investigate the conformational changes of BSA.
