摘要

Abstract

Objective To investigate the changes in lung pathology and serum level of interleukin-8 (IL-8) at different time points after smoking cessation in rats exposed to smoke for different durations, and to explore the effect of different durations of smoke exposure on lung and systemic inflammafion. Methods A total of 170 male Wistar rats were randomly divided into 30-day smoke exposure group (n =80),70-day smoke exposure group (n =80),and control group (n =10),and the two smoke exposure goups were divided into 8 sulgroups (n =10 each subgroup) according to the duration of smoking cessation (0 day,3 days,l week,2 weeks,4 weeks,6 weeks,8 weeks,and 12 weeks). The serum level of IL-8 was measured by ELISA,and the pathological changes in lung tissue were observed under light microscope.Results Compared with control group,the serum levels of IL-8 were significantly higher and the lung inflammation was more severe at each time point after smoking cessation in two smoke exposure groups (P < 0.05 ). In 30-day smoke exposure group, the serum level of IL-8 reached the peak and the lung inflammation score was the highest after 8 weeks of smoke cessation,and then the serum level of IL-8 tended to decrease and the lung inflammation was alleviated. In 70-day smoke exposure group,the serum level of IL-8 continued to increase and the lung inflammtion tended to aggravate at each time point after smoke cessaion. The serum level of IL-8 and the inflammation score were significantly higher in 70-day smoke exposure group than in 30-day smoke exposure group (P < 0.01 ). Conclusion The longer the smoking duration,the longer and more severe the lung and systematic inflammation after smoking cessation. When chronic obstructive pulmonary disease is developed,quitting smoking alone could not effectively prevent the disease process. Therefore,it is necessary to quit smoking as soon as possible.
