Abstract
Objective:Blocking the expression of Bmi-1 with shRNA intericrence techniques,the eliects of suppressing the metastasis and invasiveness of human bladder carcinoma EJ cells were investigated.Methods:The ability of migration and invasion of EJ cells were observed by Transwall and Wound closure assay.The expression of MMP-2,E-Cadhcrin and α-SMA were analyzed by RT-PCR and Western blotting.Results:In invasion experiment,the trans-membrace cell number in pGeneClipTM-Bmi-1 transfected cells was 77.0±4.8,and that in untransfected cells and in blank plasmid transfected cells was 152.1±4.3 and 155.0±6.5 rcspectively.The migration were inhibited significantly in EJ cells transfected by pGeneClipTM-Bmi-1 compared with that in EJ cells transfected by blank plasmid and untransfected cells.The expression change of MMP-2,E-Cadncrin and α-SMA in presence of TGF-β were inhibited in pGeneClipTM-Bmi-1 transfected cells.Conclusion Bmi-1 shRNA supprcsscs the migration and invasiveness of EJ cells and perform the function through blocking the TGF-β signaling pathway.关键词
