Abstract

Aim: To investigate the sensitive enhancement of cisplatin( cDDP) on tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)-mediated apoptosis in esophageal cancer EC1 cells. MethodS:The effects of TRAIL (10,50, 100,500,and 1 000 ng/L) or cDDP( 1.0,2. 0,4. 0,10. 0,and 20. 0 mg/L) on the cell proliferation of Ecl for 24,48 and 72 h were observed by MTT. EC1 cells were allocated into 4 groups and treated with 100 mg/L TRAIL or 100 mg/L cDDP or combined use of the 2 drugs for 48 h, and the cells not being treated were control. The apoptosis and cell cycles of EC1 were measured by flow cytometry. ResultS;TRAIL did not apparently influence the proliferation of EC1 cells( P > 0. 05) , however, CDDP inhibited the proliferation of EC1 cells(P <0.05) after 24,48 and 72 h treatment. TRAIL had no influence on the cell apoptosis rate of EC1 ( FTRAlL. =5. 836, P =0. 250) , while cDDP could increase the cell apoptosis rate (FcDDP =20. 763,P =0.041). Furthermore, the cell apoptosis rate was greatly increased when the 2 drugs were combined (Fbath, = 57.422,P<0.001). TRAIL or cDDP single had no effects on cell cycle, while combined use of the 2 drugs decreased the percentages of Go/G1 and S phase cells and increased the percentage of G2/M phase cells( P < 0. 05). Conclusion ;cDDP has sensitive enhancement on TRAIL-mediated apoptosis in EC1 cells. TRAIL in combination with cDDP induces EC1 apoptosis and cell cycle to arrest at G2/M phase.
