云南2株H4N6亚型禽流感病毒株的分离与鉴定
Isolation and identification of 2 strains of H4N6 avian influenza virus in Yunnan Province

Abstract
Two laryngotracheal samples were collected and demonstrated for the infection of avian influenza virus ( AIV) by the RT-PCR amplification of M gene. The samples were further verified negative for the H9, H5 subtype AIV by the established PCR methods. The 11-day-old chicken embryonated eggs were used to cultivate the virus, and then the allantoic fluid was collected for the hemagglutination and he-magglutination inhibition assays. The positive blood sera of H5, H7 and H9 subtype AIV and Newcastle disease virus could not inhibit hemagglutination of the allantoic fluid. Homology analysis of HA gene revealed that sequence similarities varied from 34. 9％ to 87. 3％ with the different HA subtypes, and showed the highest identity with H4 subtype, 87. 3％. Homology analysis of NA gene revealed that sequence similarities varied from 47. 7％ to 90. 8％ with the different NA subtypes, and showed the highest identity with N6 subtype, 90. 8％. The results demonstrated that the two unknown virus strains are H4N6 subtype of AIV.关键词
