摘要

Abstract

Objective: To investigate the expression patterns and the roles of eNOS and nNOS during development of the embryonic mouse kidney.Methods: The mouse were divided into six groups of embryonic day 13 (E13), E14, E15, E16, E18 and postnatal day (PO) according to embryo development.The expression of eNOS and nNOS was examined by immunohistochemical and analyzed in a cell image analysis system.The contents of eNOS and nNOS were measured by Western blot.Results: (1)The expression of eNOS was identified in the nephrogenic zone from El4.At El6, it was clearly seen in proximal tubules.By PO, it was expressed in distal tubules and collecting ducts, but could not be seen in macula densa.(2)The nNOS was expressed in ureteric bud (UB) at E13 d.The staining was stronger in distal tubules but more weakly in the proximal tubules at El6 d.The positive expression of nNOS could be identified in macula densa.The results of image analysis and Western blot indicated that the content of eNOS and nNOS gradually increased from E14 onward, and reached to peak at PO.Conclusion: The expression of eNOS and nNOS in the developing kidney of embryonic mouse has temporospatial sequence.They may play an important role in the development of the embryonic mouse kidney.
