青蒿琥酯对乳腺癌细胞株MDA-MB-231生长的影响

Abstract
Obective To study the. inflence of artesunate on proliferation of breast cancer MDA-MB-231 cells and it's possible mechanism. Methods Use different concentrations( 0、25、50 、1OOμg/ml) of Artesunate to treat the MDA-MB-231 cells. The MTT assay was used to detect the growth of cells ; the cell proliferation index was measured by microscope and the cell cycle( mainly the ratio of G2/M phase and apoptosis cells) was analyed by flow cytometry. Results The results of MTT assay showed Artesunate could inhibit the proliferation of MDA-MB-231 cells , with the increase of drug concentration and time treated by drugs, the inhibitory action also raised. When compared the experimental groups with the control grop , there are sighificant differences analysed by the statistical software( P ＜ 0. 05 ) . The ratio of cells in G2 phase and/or mitotic phase and the proportion of apoptotic cells increased significantly after treated by Artesunate analyzed by the results of cell proliferation index and the flow cytometry. when compared with the control group, the difference has statistical meaning( P ＜ 0. 05) . Conclusion Artesunate can inhibit the proliferation of MDA-MB-231 cells by prohibiting their division and induce apoptosis, Artesunate can be used as a potential drug to treat hreast cancer.关键词
