甘南藏系绵羊DGAT1基因16-17外显子多态性分析
Polymorphism at 16-17 Exon of DGAT1 Gene in Tibetan Sheep of Gannan Tibetan Prefecture
摘要
Abstract
The aim of the study was to investigate the single nucleotide polymorphisms(SNPs) at 16-17 exon of DGAT1 (diacylglycerol acyltransferasel) gene in Tibetan sheep breeds and Tan sheep,and provide a foundation for studying the relationship of DGAT1 gene with some growth and carcass traits,the genetic differentiation of Tibetan sheep breeds and the genetic relationship between Tibetan sheep breeds and Tan sheep. A total of 501 sheeps were used for investigating.SNP was detected by comparing sequences of PCR products,and the restriction fragment length polymorphism (RFLP) technique was adopted for genotyping. The result showed that: ①There were two alleles T and C,and allele C was predominant. Three genotypes TT, TC and CC for SNP at 16-17 exon of DGAT1 gene in Tibetan sheep breeds were discovered,and the predominant genotypes was CC. ②Chi-square test indicated that genotype of 16-17 exon at Alu Ⅰ locus of DGAT1 gene in Oula sheep was in Hardy-Weinberg equilibrium (P>0. 05),Ganjia sheep and Qiaoke sheep were not in Hardy-Weinberg equilibrium (0.01<P<0. 05).③The compatability test of genotype distribution at this mutated locus showed that there was significant difference between Tan sheep and the Tibetan sheep breeds. ④Polymorphism information content at 16-17 exon of DGAT1 gene showed that the Tibetan sheep breeds are moderate polymorphism. The result indicate that there are less hereditary difference among the three Tibetan sheep breeds(P>0. 05),but there are obvious hereditary difference between Tibetan sheep breads and the Tan sheep(0. 01<P<0. 05)in SNP at 16-17 exon of DGAT1 gene.关键词
