d-δ-三烯生育酚对人鼻咽癌CNE2细胞株增殖和凋亡的影响
Effect of d-δ-tocotrienols on the proliferation and apoptosis of nasopharyngeal carcinoma CNE2 cell line
摘要
Abstract
Objective To investigate effect of d-δ-tocotrienols on proliferation and apoptosis of nasopharyngeal carcinoma.Methods The experiment was carried out with control group and experimental group which included three different concentration of dδ-tocotrienols(40,50,60 μmol/L).Cytomorphology was analyzed by using Hoechst 33 258 fluorescent staining and microscopy.MTT experiment and flow cytometry were used to analyze the effect of d-δ-tocotrienols on proliferation,apoptosis and cell cycle of nasopharyngeal carcinoma cell line CNE2.Results After treated CNE2 cells with d-δ-tocotrienols for 48h, cell growth suppression rate rose obviously with the increase of d-δ-tocotrienols concentration, respectively be (27.71± 1.01)% , (57.09±0.56)% and (95.04±0.17)% ,the differences were significant when compared with the control group (P<0.01).Apoptotic cells with nucleus and cytoplasma compact dyeing or thick strip massive fluorescence increased obviously, the apoptotic cell rates were (9.18±1.86)% , (34.97±1.01) % and (52.97±2.77)% respectively in experimental group, compared with the control group (1.58±0.48)% ,cell apoptosis rate significantly increased,the differences were significantly (P<0.01).The percentage of S phase cells were (25.40±1.93)% , (35.83±3.74)% and ( 47.13±2.49)% respectively in experimental group, cells were stopped at S phase,compared with control group (19.63±1.46)% ,the differences were statistically significant (P<0.01).Conclusion d-δ-atocotrien-ols has biological activity of anti-nasopharyngeal carcinoma CNE2 cell line.关键词
