体外诱导的骨髓源肝细胞样细胞移植治疗肝功能衰竭的实验研究
Study on the therapy of hepatic failure using hepatocyte-like cells derived from bone marrow transplantation

Abstract
Objective To explore the therapeutic effect of acute hepatic failure by transplanting hepatocyte-like cells derived from bone marrow.Methods BMSCs were differentiated in vitro.RTPCR and flow cytometry (FCM) were performed to examine the cellular differentiated states by exploring the expression of BST-1 and albumin.Rats with acute hepatic failure model were randomly divided into 2 groups: experimental group(hepatocyte-like cells transplantation) and control group.The levels of biochemistry,pathological morphology and survival state analysis were used to evaluate the ability of hepatocyte-like cells derived from bone marrow to improve the liver functions.Results Hepatocyte-Iike cells expressed the gene of albumin, but the gene of BST-1 disappeared after the induction for 20 days.The result of FCM showed that hepatocyte-like cells expressed albumin too.Hepatocyte-like cells transplantation could lower the level of AST, ALT and TBIL,enhanced the ability to synthesizing ablumin,and improved the liver functions and survival rate of acute hepatic failure rats remarkably.Conclusion Hepatocyte-like cells derived from bone marrow can improve hepatic function,which can be used for a perfect cell source of HCT.关键词
