摘要

Abstract

[ Objective ] This study aimed to establish in vitro conservation system for fruit tree Malus baccata. [ Method ] With the north common fruit tree Mains baccata as the tested materials, they were cultured on the MS basic medium added by mannitol with different concn. at 0, 5, 10, 15, 20, 25 g/L for in vitro conservation at normal temperature and low temperature(5℃)resp. to study the effect of mannitol concn. and the temperature condition on the conservation of test tube seedlings of M. baccata. [Result] Under normal temperature, adding the mannitol at 15 g/L into the MS medium could get better conservation and the test tube seedlings could be conserved for 9 months, with the conservation time being close to CK that had the conservation for 8 months. Under low temperature, the conservation cycle of the test tube seedlings was increased and adding the mannitol at 10 and 15 g/L into the MS medium could get better conservation, with the conservation cycle being delayed to 20 months. The observation on the test tube seedlings which were conserved for 16 months under normal temperature and low temperature and then were made for subculture and rooting culture, it was found that their biological characteristics were no obvious change. [Conclusion] The optimum mannitol concn. for M. baccata concentration was 15 g/L at the normal temperature and that was 10 - 15 g/L at the low temperature.
