Abstract

[Objective] The purpose was to enhance the acclimation efficiency of ex vitro rooting of Populus bachofenii × P. pyramidalis ROZ. [Method] With the clonal tissue cultured micro cuttings of P. bachofenii × P. pyramidalis as materials, the correlation between micro-environmental factors and its ex vitro rooting rate and roots quality was studied through comparative experiment. [Result] The rooting effect of micro cuttings on the medium 25% peat + 30% vermiculite + 45% perlite was best and its subordinate function value, rooting rate, root number, root length and partial root rate were 0. 857, 90% ,11.6 per plant, 2.25 cm and 11 % resp. The rooting effect of micro cuttings treated with IBA was best and the subordinate function value, rooting rate, average root number, average root length and partial root rate were 0.785, 93.33% , 6.12 per plant, 1.15 cm and 13.67% resp. in the treatment with 300 mg/L IBA. The rooting effect of micro cuttings was best under 20 ℃ and its subordinate function value, rooting rate, root number, root length and partial root rate were 0.820, 100.00% , 7.80 per plant, 1. 17 cm and 15.33% resp. The rooting effect of micro cuttings treated with light intensity of 75 μmol/(m~2·s) was best and its subordinate function value, rooting rate, root number, root length and partial root rate were 0.686, 100.00% , 8.76 per plant, 2.08 cm and 38.00% resp. The rooting effect of micro cuttings on the medium with water content of 50% was best and its subordinate function value, rooting rate, root number, root length and partial root rate were 0.894, 100.00% , 7.73 per plant, 1.75 cm and 13. 33% resp. [Conclusion] The study laid a foundation for establishing the rapid and effective micro-propagation technical system of P. bachofenii × P. pyramidalis.
