摘要

Abstract

The microRNAs (miRNAs) are a newly identified class of small non-coding RNA,which regulate post-transcrip-tional gene expressing in eukaryote. Most miRNAs are predicted through the computer, the presentence/absence and the spatial/temporal of miRNAs still unclear. It is important for the establishment of efficient, sensitive and specific detection methods to detect miRNAs in different organism or period. In this study,the protocol derived from Lau et al. and Shi et al. was used to detect miR156a,predicted and released by Sanger Institute,in old or young leaf,root,and shoot for verifying this method. By comparison,this method is low cost, simple,fast, and sensitive, common for primer of RT-PCR, which could i-dentify a number of miRNAs in the same organism and compare miRNAs from different organism. Furthermore, this method need less RNA,in comparison with Northern Blot and Microarray.
