Effect of immune cells mainly composed of dendritic cells on peripheral blood levels of IFN-γ and TNF-α in patients with chronic aplastic anemia

Abstract
Objective To study the changes of interferon - γ (IFN - γ) , tumor necrosis factor - α (TNF - α) in peripheral blood (PB) of patients with chronic aplastic anemia (cAA) treated with immune cells mainly composed of dendritic cells(DC). Methods The expression of intracellular IFN -γ and TNF - α in PB CD3~+ and CD8~+ T cells of 14 normal controls were detect by flow cytometry, and the expression of plasma IFN - γ and TNF - α was measured with ELJSA. Fourteen primary patients with cAA were treated with immune cells mainly composed of DC for 10 months. The expression of IFN - γ and TNF - α in PB CD3~+ and CD8~+ T cells was detect by flow cytometry and their levels in peripheral blood plasma were measuered by ELISA before and 10 months after treatment. Results Elevated levels of IFN - γ and TNF - α were found in blood plasma and CD3~+ and CD8~+ T lymphocytes of AA patients compared to normal controls (P <0. 05). After 10 months, there was a significant decline in plasma IFN - γ and TNF - α, intracellular IFN - γ, TNF - α in CD3~+ and CD8~+ T cells in PB compared to those before treatment (P < 0. 05 ) ; there were no significant differences of these cytokines compared to normal controls (P > 0. 05). Conclusion Immune cells therapy may induce he-matological remission by suppressing the elevated levels of IFN - γ and TNF - α in AA PB CD3~+ and CD8~+ T cells and plasma.关键词
