山楂果实不同组织RNA提取方法比较及cDNA-SRAP分析体系的建立
Comparison of methods used for RNA extraction from different tissues of hawthorn fruit and establishment of cDNA-SRAP system

Abstract
Total RNA from different tissues of soft-core hawthorn fruit was extracted with two improved methods: Trizol and Hot-borate step.The quality and the content of total RNA was tested by nucleic acid/protein assay and gel electrophoresis.The result showed that the improved Trizol method was so effective to produce 60.3 μg·g~(-1) and 46.2 μg·g~(-1) high quality total RNA from the pulp and the core ,TheA_(260)/A_(230)of yield RNA was more than 2.0 while A_(260)/A_(230) was 1.92 and 1.84 separately, but it was difficult to extract RNA from seed.Compared with the improved Trizol method,the improved hot-borate step was effective to produce more high quality total RNA from the hawthorn pulp,core and seed.The RNA extracted from the hawthorn with the improved hot-borate method couM be used in cDNA-SRAP, the clear bands were obtained which was suitable for common molecular biological researches.关键词
