流感病毒在诱导A549细胞凋亡过程中对SIRT1和P53蛋白的影响
Influences of Influenza Virus on SIRT1 and p53 in Inducing Apoptosis of Human A549 Cells
摘要
Abstract
To study the regulation of SIRT1 expression in influenza virus-induced apoptosis in A549, we observed apoptosis of A549 cell by flow cytometry during influenza virus-induced apoptosis, and detected protein expression of SIRT1, p53, Bax and Bcl-2 by western blot analysis. The results showed that A549 cells exhibited typical characteristics of apoptosis, and percentage of cell death was obviously increased after the treatment with 1 000 TCID_(50)/mL influenza virus. SIRT1 protein expression in A549 cells was decreased by the infection of influenza virus. Meanwhile, phosphorylation p53 expression and the ratio of Bax/Bcl-2 protein expression were both elevated. Taken together, influenza virus decreased SIRT1 expression in A549 cells, which strengthened apoptosis action of Bax and p53 protein.关键词
