摘要

Abstract

OBJECTIVE: To explore the effect of 3, 4-Dihydroxyacetophenone (DHAP) on the expression of Toll-like receptor 4 (TLR4) in activated mouse vascular smooth muscle cells (VSMC), endothelial cells (EC) and macrophages (MC).METHODS: Normal thoracic aortas were obtained for primary culture of VSMC and EC and serial subcultivation of RAW264.7 cell lines. The cells were divided into control group, LPS group,simvastatin group, DHAP high dose group and DHAP low dose group. Total mRNA and protein content of each group were analyzed by RT-PCR and Western blot, respectively. RESUL-TS: The expression of TLR4 mRNA and protein content of VSMC, EC and MC in DHAP high dose group were significantly lower than in LPS group(P<0.01).CONCLUSION: DHAP could down-regulate the LPS-activated expression of TLR4 mRNA and protein in VSMC, EC and MC of mice, and TLR4 path might be the anti-inflammatory mechanisms of DHAP.
