HPLC法测定不同生长期粤北产山药中腺苷的含量
Determination of the content of adenosine in Dioscorea opposita Thunb.of different growth period growing in the North of Guangdong by HPLC

Abstract
Objective To determine the content of adenosine in Dioscorea opposita Thunb. Of different growth period growing in the North of Guangdong by a high performance liquid chromatography( HPLC) method in order to confirm the best harvesting time. Methods The operation was performed on a Discovery C,8 column (5 ujn,250 mm x4. 6 mm) using acetonitrile-water (5: 95) as the mobile phase at the flow rate of 1.0 mL ? Min "'. The detection wavelength was set at 260 nm. Results The calibration curve of adenosine displayed ideal linearity over the range of 0.092 4 -0.554 4 u,g(r = 0.999 7) ,and the average recovery was 98. 8% ( n = 6). The content of adenosine increased gradually from the first to the twelfth month and reached its peak in the the twelfth month. Conclusion The method established could be used for quality control of Dioscorea opposita Thunb. Growing in the North of Guangdong due to its convenience, good reproducibility and high accuracy,and the best harvesting time was the twelfth month.关键词
