Methylation status of promoter of SPARC in epithelial ovarian cancer.

Abstract
Objective To study the role of promoter hypermethylation of SPARC gene in the course of tumorigenesis and development in epithelial ovarian carcinoma. Methods The methylation status of promoter region SPARC was assessed by methylation-specific polymerase chain reaction (MSP) in formalin fixed,paraffin embedded tissues from 40 pithelial o-varian cancer patients,30 benign ovarian tumor patients and 10 normal ovarian patients. Results The methylation frequency of SPARC was65.0% (26/40) ,26.7% (8/30) and 20% (2/10) in ovarian cancers, benign ovarian rumors and normal ovarian tissues respectively. Between the different organization type the hypermethylation of SPARC gene was more frequently encountered in ovarian cancers than in tumor and normal tissues (P < 0. 05 ). There was no difference between tumors and normal tissues,and also between the different organizationtype(P >0. 05). The frequency of promoter hypermethylation of SPARC was significantly lower in epithelial ovarian cancers of stage I and II than that in stage 11 and (P < 0. 05), hydroperitoneum group was higher than that without hydroperitoneum ( P < 0.05). But there was no difference between lymph node metastasis and no lymph node metastasis (P > 0.05 ). Conclusion The methylation of promoter hypermethylation of SPARC gene has been postulated to play an important role in the development of ovarian cancer. This methylation correlates with histopathologic grade and clinical stage of epithelial ovarian cancer.关键词
