Effects of genistein on the level of oxidative stress in rat genioglossus muscle under chronic intermittent hypoxia condition.

Abstract
Objective: This study aims to investigate the effects of genistein on the oxidative stress level and anti-oxidant enzyme activity in genioglossus of rats under chronic intermittent hypoxia condition.Method: Fifty female SD rats (8 weeks old) were randomly divided into five groups 1 week after ovariectomy: the normal control group, the chronic intermittent hypoxia (CIH) group, the CIH plus low dose,moderate dose,and high dose genistein groups.Rats in the latter four groups were exposure to CIH for 8 h/d for 5 weeks.DCFH-DA fluorescent probe,thiobarbituric acid assay,and substrate catalysis assay were used to detect the level of ROS and MDA and the activity of anti-oxidant enzyme in five groups.Resuit: The level of ROS and MDA was increased by CIH and reduced by genistein (P <0.05).The activity of SOD, CAT, and GPx was decreased by CIH (P <0.05).Different doses of genistein recovered the activity of CAT and GPx to normal level,while only high dose of genistein recovered the activity of SOD to normal level (P <0.05).Conclusion: CIH exposure increased the level of oxidative stress in genioglossus muscle, while genistein treatment could reduce the level of oxidative stress through enhancement of anti-oxidant enzymatic activity.关键词
