荧光法测定同型半胱氨酸的方法学评价

Abstract
Objective To evaluate the features of total Homocysteine (Hcy) assay using novel biochemical assay. Methods Performance characteristics such as precision,linearity,interference testing were evaluated respectively according to EP5-A,EP6-A and EP7-A issued by the National Committee on Clinical Laboratory Standards(NCCLS). Results The CVs of within-run and total precisions at high concentration leveI(Hcy 20.0 μmol/L) were 1. 88% and 3. 71% respectively,CVs of within-run and total precisions at low concentration level(Hcy 10. 0 μmol/L) were 2. 76% and 3. 98% respecrively. The linear range was 0~60. 0 μmol/L. When the concentration of bilirubin was at 500 μmol/L or triacylglyceride at 15. 0 mmol/L or vitamin C at 10. 0 mmol/L or hemoglobin at 16. 0 g/L produced interference deviation was less than 5 %. Correlation bewteen results obtainedby this method and hplc method was well,decisive coefficient (r2) =0. 976 1. Conclusion Performance of precision,linearity and interference testing of the homocysteine assay kit produced by XingNen Company could fulfill the requirements of clinical laboratory.关键词
