养肝益水颗粒对自发性高血压大鼠肾脏Smad2、Smad3、Smad7表达的影响
Effects of Yanggan Yishui Granules on Expression of Smad2, Smad3, and Smad7 in Renal Tissues of Spontaneously Hypertensive Rats

Abstract
Objective To observe the effects of Yanggan Yishui Granules (YGYSG) on the expression levels of Smad2 , Smad3 ,and Smad7 in the renal tissues of spontaneously hypertensive rats( SHR) , in an effort to elucidate the mechanism of YGYSG in protecting the kidney. Methods Fifty 12-week-old male SHRs were averagely randomized into model group, Benazepril (1. 8 mg/kg daily) treated group, low (5. 4 g/kg daily)-, and high (27 g/kg daily)-dose YGYSG treated groups; and another ten Wistar-Kyoto (WKY) rats were used as normal controls. The rats in model group and control group were treated with normal saline, once daily. Six weeks later, the expression levels of smad2, smad3, and smad7 in the renal tissues were assayed using immunohistochemistry. Results Compared with those of the control group, the expression levels of Smad2, and Smad3 in the renal tissues were significantly increased in the model group (P< 0. 01) , while the expression levels of Smad7 were significantly reduced (P<0. 01). Compared with those of the model group, the expression levels of Smad2, and Smad3 in the renal tissues were significantly reduced (P<0. 01), while the expression levels of Smad7 were significantly increased in the three treated groups (P<0. 01). There was no significant difference in the expression levels of Smad2, Smad3, and Smad7 between low-dose YGYSG treated group and Benazepril treated group (P>0. 05) ; while the difference was significant between high-dose YGYSG treated group and low-dose YGYSG treated group (P< 0. 01), and between high-dose YGYSG treated group and Benazepril treated group ( P < 0. 05 ). Conclusion YGYSG can protect the kidney from fibrosis by blocking transforming growth factor-transforming growth factor receptor-Smads signaling pathway via down regulating the expression levels of Smad2 , and Smad3, and up regulating the expression levels of Smad7 in the renal tissues.关键词
