Influence of SOCS-1 on TNF-α-induced renal tubular epithelial cells apoptosis

Abstract
Aim To investigate the effects of suppressor of cytokine signaling-l ( SOCS-1 ) on tumor necrosis factor alpha ( TNF-α ) induced apoptotic cell death and activation of JAK/STAT in cultured human renal tubular epithelial cells ( HKC ). Methods Stable transfections of HKC with pCR3. I vector and pCR3. 11SOCS-I were performed with Lipofectamine 2000, and cells were selected with geneticin. Cells were stimulated with TNF-a( 20 μg · L-1 ). The protein expressions of SOCS-I. Bax, Bcl-2. Cleaved caspase-3,Caspase-3 Janus kinase 2/ signal transducer and activator of transcription I ( JAK2, STATI ), p-JAK2 and p-STATI were observed by Western blot. Apoptosis rate of HKC induced by TNF-a was analyzed by flow cytormetry ( FCM ). DeadEndrM Colorimetric TUNEL System detection kit were used to observe the number of apoptotic cells. Results Compared with control group, the number of cell apoptosis significantly increased, meanwhile, the expression levels of Cleaved caspase-3 protein and Bax/Bcl-2 were significantly increased in HKC with TNF-a stimulation. Overexpression of SOCS-1 inhibited TNF-a-induced cell apoptosis, activation of JAK2 and STATl,and expression of Cleaved caspase-3 protein, as well as the ratio of Bax/Bcl-2. Conclusion Overexpression of SOCS-I may inhibit TNF-α-induced renal tubular epithelial apoptosis partly through blocking activation of JAK/STAT.关键词
