游动放线菌中orf20基因敲除对雷莫拉宁合成的影响
Effects of gene disruption of orf20 on ramoplanin production in Actinoplanes sp.
摘要
Abstract
The orf20 in the ramoplanin biosynthesis gene cluster shows high homology to the known halogenases . An inframe knock-out plasmid pSM-3 was transformed into Actinoplanessp . ATCC 33076 to delete the orf20 gene fragm entencoding 64 am inoacids. A representative orf20 mutant was designated SIPI-A .2001 dorf20 (△orf20 ) .Absence of ramoplanin and its analogs production from the rsulting exconjugants suggested that the protein coded by orf20 was a key enzyme in the ram oplanin biosynthesis pathway , and it might be have other funcions except halogenation .关键词
