The inhibitory effects and induction of fibrinolytic protein from Eupolyphaga sinensis on the apoptosis of tumer S180 and H22 cells in mice

Abstract
Aim To study the induction effects of fibrinolytic proteins isolated from Eupolyphaga sinensis ( EFT ) on the apoptosis of tumer S180 and H22 cells in mice and its mechanisms. Methods The molecularweight of EFP was detected by sulfate-polyacrylamide gel electrophoresis analysis. The fibrin plate assay was used to test the fibrinolytic activity of EFP. Tumor bearing mouse models ( solid and ascites tumors of S180 and H22 ) were constructed by injecting tumor cells. Cell apoptotic rate was detected by flow cytometry following annexin-V/PI staining. Caspase-3 levels of solid tumors S180 and H22 were measured by spectrophotometry. Effects of EFP on telomerase activity of H22 and S180 ascites tumor cells was detected by telomeric repeat amplification protocol ( TRAP )-silver staining. Results EFP had the effects of inducing apoptosis, raising the level of Caspase-3 of solide tumer S180 and H22 . and decreasing the telomerase activity of ascites tumors of S180 and H22. Conclusion EFP can induce cell apoptosis of S180 and H22 cells.关键词
