巴西橡胶树树根组织培养外植体消毒研究
Studies on the Explant Disinfection in the Root Tissue Culture of Hevea brasiliensis
摘要
Abstract
[ Objective ] The aim was to look for an effective way of decreasing explant contamination in the root tissue culture of Hevea brasiliensis. [ Method]The root tissue culture of H. brasiliensis was carired out taking news roots after treatment for 30 d as explants,and explants was disinfected by using 1 g/L carbendazim,0. 1% HgCl2 and Yipeilong with different concentration. [ Result] Explants were treated by 1 g/L carbendazim before conventional disinfection and disinfected by 75% ethyl for 30 s and 0.1% HgCl2 for 6 min,successively, finally explants were inoculated in solid callus induction medium with 0. 1% Yipeilong and cultured for 30 d when the contamination rate of explants decreased to 44.59% and its survival rate was up to 25.60%. [ Conclusion] The study could provide theoretical foundation for the decrease of explant contamination in the root tissue culture of H. brasiliensis.关键词
