紫外诱变原生质体选育低温碱性脂肪酶高产菌株
Breeding of cold-adapted and alkaline lipase producing mutants by protoplast mutagenesis of ultraviolet irradiation
摘要
Abstract
The cold-adapted and alkaline lipase producing strain Acinetobacter johnsonii LP28 was selected as the parent strain and the protoplast was prepared with ED TA and lysozyme . The corresponding values usad were 0.15mg/mL lysozyme , at 37℃ for 45 min . The rate of cell oonverted to protoplast was 90% and the regeneration efficiency was about 0.9% . Acinetobactrr johnsonii LP28 was induced by protoplast mutagenesis of ultraviolet irradiation and the mutant P169 was obtained . Its lipase activity was as high as 5.16 U/m L , increased by 2.19 tmes in comparison with that of the Acinetobacter johnsonii LP28 .关键词
